After entering the cave by boat, dock on the beach to the right. Here grab a bomb, then turn around and shoot a sigil (purple) arrow at the vines on the wall. Now throw the bomb at the sigil to get rid of the vines. Now shoot 3 sigil arrows that are touching each other as well as all 3 runes. Now hit the middle sigil with a bomb to create a chain reaction and activate all 3 bells to open the chest that can be found on the other side of the rocks to the right of the bomb.
